بررسی اثر آنتی‌اکسیدانت‌ رزوراترول بر پارامترهای بیوشیمیایی اسپرم و بیان ژن‌های کاسپاز3 و HSP70 افراد نابارور آستنواسپرمی تحت شرایط انجماد

چکیده

مقدمه: یکی از مهم­ترین اقدامات به­ منظور افزایش کیفیت اسپرم و در نتیجه موفقیت فناوری کمک ‌زادآوری (ART)، محافظت از اسپرم در برابر واکنش ­های اکسیداتیو در حین انجماد می ­باشد. زوراترول یک ترکیب آنتی­اکسیدانی است که با از بین بردن رادیکال ­های آزاد، از بدن در برابر بیماری­های مزمن محافظت می ­نماید. هدف از  مطالعه حاضر تعیین اثر رزوراترول بر پارامترهای عملکردی اسپرم انسان و ارزیابی آپوپتوز و بیان ژن­های HSP70  و Caspase3 پس از انجماد  می ­باشد.
مواد و روش­ها: در این مطالعه از 40 مرد مراجعه کننده به مرکز ناباروری ACECR قم که همگی معیارهای ورود به مطالعه را دارا بودند به عنوان جامعه هدف استفاده شد. جامعه هدف به دو گروه مردان با معیار WHO طبیعی و گروه دوم افراد مبتلا به استنوتراسپرمی تقسیم شدند. پس از نمونه­ گیری اسپرم، اسپرم ­ها با غلظت ­های مختلف رزوراترول منجمد شدند.
نمونه­های منی از نظر پارامترهای اسپرم شامل غلظت، مورفولوژی، تحرک، زنده­ ماندن، گونه­ های اکسیژن واکنش ­پذیر(ROS) ، آپوپتوز و بیان ژن­های HSP70 و کاسپاز 3 مورد بررسی قرار گرفتند.
یافته­ ها: رزوراترول 02/0 و 05/0 میلی­ مولار به­ طور قابل­ توجهی تحرک اسپرم را افزایش داد (P، اما در مقایسه با گروه شاهد با رزوراترول 25/0 و 1/0 میلی­ مولار کاهش یافت (P. به­جز رزوراترول  25/0 میلی­مولار، همه دوزهای رزوراترول به­ویژه 05/0 میلی­ مولار باعث افزایش اسپرم شد (05/0P˂). تمام دوزهای رزوراترول به­ویژه 02/0 میلی­مولار و 05/0 میلی­ مولار باعث کاهش آپوپتوز و بیان ژن­های HSP70 و Caspase3 گردید.
نتیجه­ گیری: به­ طور کلی، مکمل رزوراترول قبل از انجماد به­ طور چشمگیری کیفیت اسپرم را افزایش داد و باعث کاهش عوارض جانبی شوک انجماد گردید.
